I'm a long-time "Sluggite" (Sluggy Freelance being among the first ever web-comics and one of only two that I know of to have made it to its 10th birthday). SF combines silly gags with complex story arcs that unfold over the course of multiple years.
My other steady favorite is Diesel Sweeties, an exemplar of reconstructivism.
I'm also a fan of the communal dream journal, Slow Wave.
I have other favorites but most of them are defunct.
I don't laugh out loud at my computer monitor very often, but that one usually does the trick.

Ah, jeez, I almost forgot! This one went the way of the dodo several years ago, but the archives are still there. I think I may have wet myself more than once reading these in college. The crazy thing is that, every now and again, you'll stumble upon one that isn't funny at all... more disturbing (and genuinely so, especially in contrast to the rest) than anything. Brilliant, in either case:
I actually prefer Drew's other comic Married to the Sea over Toothpaste for Dinner. I've never been able to get past the lumpy slouching-dude style of TfD. He also makes some funny videos. Unfortunately, I don't read his comics very often as he doesn't have an RSS feed :( (Or at least, I've never found one).
Some that I do read regularly:
Scary Go Round : easily my favorite webcomic at the moment. I love John Allison's art style. I really dug his Illustrator drawings and was afraid I wouldn't dig it as much when he switched back to hand-drawn, but he makes the best character expressions.
Questionable Content - indie-music tinged, funny "life" or relationship comic.
The Non-Adventures of Wonderella - twisted sort of postmodern "superhero" comic
Sinfest - hmmm.. not sure how to describe Sinfest. It's newspaper-stripy, maybe think of Calvin grew up (a few years at least), replaced Hobbes with a talking pig, did poetry jams, chased his muse, and interacting with God, the Devil, and Buddah.
Count Your Sheep - about a girl, her mom, and *their* imaginary friend, a sheep. I used to LOVE CYS, but it's been losing me of late...
Side note: There is a new comic book shop opening next to Rave movie theater up by Polaris. It's a huge store space so we'll see how long that lasts.

At midnight moving on to Wednesday my new webcomic Guest Artist Week will debut. It's communally written and drawn so if any of you would like to contribute scripts and/or art, please let me know. Right now we have extra scripts, but we're looking for more clippable art, full strips, or people willing to compile strips from the art and scripts we already have.
Basically, the idea is it's a strip where the regular artist and writer are ALWAYS on vacation. Here for more...
Sounds like a great concept! Can't wait to see more of the strip.
Penny Arcade is a classic.
And Tailsteak is pretty clever. I like his 20 Questions with a penny.
